Evidence suggests that insulin resistance plays an important role in developing diabetes complications. The association between insulin resistance and pain perception is less well understood. This study aimed to investigate the effects of peripheral insulin deficiency on pain pathways in the brain. Diabetes was induced in 60 male rats with streptozotocin (STZ). Insulin was injected into the left ventricle of the brain by intracerebroventricular (ICV) injection, then pain was induced by subcutaneous injection of 2.5% formalin. Samples were collected at 4 weeks after STZ injection. Dopamine (DA), serotonin, reactive oxygen species (ROS), and mitochondrial glutathione (mGSH) were measured by ELISA, and gene factors were assessed by RT-qPCR. In diabetic rats, the levels of DA, serotonin, and mGSH decreased in the nuclei of the thalamus, raphe magnus, and periaqueductal gray, and the levels of ROS increased. In addition, the levels of expression of the neuron-specific enolase and receptor for advanced glycation end genes increased, but the expression of glial fibrillary acidic protein expression was reduced. These results support the findings that insulin has an analgesic effect in non-diabetic rats, as demonstrated by the formalin test. ICV injection of insulin reduces pain sensation, but this was not observed in diabetic rats, which may be due to cell damage ameliorated by insulin.

BACKGROUND AND OBJECTIVES: This study aimed to evaluate the volume, the concentration of steroid hormones, and biochemical composition of the foetal fluids at different gestational ages in dogs and cats. METHODS: Following the ovariohysterectomy, the allantoic and amniotic fluid samples were collected from pregnant bitches and queens and were assigned to different groups according to their gestational age. RESULTS: The canine and feline allantoic fluid volume increased during pregnancy, reached its maximum values on days 40-49 and then decreased. The canine and feline amniotic fluid volume increased steadily by the last days of pregnancy. In spite of significant changes of sex hormones in the foetal fluids, their concentration and ratios were not significantly different between male and female fetuses. The canine amniotic cortisol concentration increased until days 40-49 and decreased significantly afterwards. The maximum cortisol concentrations in the feline allantoic and amniotic fluids were observed on days 50-60 and 40-49, respectively. During the canine pregnancy, the concentrations of calcium, phosphorus, chloride, sodium, triglyceride, cholesterol, total protein, albumin and the activities of aminotransferase (AST), alkaline phosphatase (ALP), amylase and gamma-glutamyl transferase (GGT) in the amniotic fluid were higher than the allantoic fluid. The magnesium, potassium, lactate dehydrogenase (LDH) activity, creatine and lipase were higher in the allantoic fluid. In the feline allantoic fluid, potassium, magnesium, phosphorus, creatinine, albumin and glucose concentrations and the activities of creatine kinase (CK), GGT, LDH and lipase were higher. The ALP, AST activities, sodium and calcium concentrations were higher in the amniotic fluid (p < 0.05). CONCLUSION: Volume of foetal fluids was determined in dogs and cats. Concentration of sex hormones did not different between male and female fetuses.

One-humped camels (Camelus dromedarius) exhibit remarkable adaptability to harsh desert environments through various physiological adaptations. This study aimed to assess variations and reference values of Heat-shock proteins (HSPs), physiological parameters, mineral concentrations, total antioxidant capacity (TAC), and malondialdehyde (MDA) in 90 healthy female one-humped camels from Zabol's outskirts in Iran. The objective was to understand how these camels adapt to heat stress. Blood samples were collected from camels located at five geographical regions and analyzed using standard kits and methods. Reference intervals for heat-shock protein 30 (HSP30), heat-shock protein 40 (HSP40), heat-shock protein 70 (HSP70), and heat-shock protein 90 (HSP90) were determined using the reference value advisor (RVA). The study found significant differences among different regions for HSPs (P < 0.05), MDA (P = 0.021), and TAC (P = 0.042) levels, indicating variations in adaptation mechanisms. However, no notable differences were observed for other measured parameters between these regions. There were no significant differences observed in the evaluated parameters between the age categories of > 36 months and < 36 months. The positive correlation between HSPs and MDA levels (ranging from 0.754 to 0.884) suggests that the synthesis of HSPs is triggered as a response to oxidative stress caused by an imbalance between the production of reactive oxygen species (ROS) and the body's antioxidant defenses. This oxidative stress, in turn, is a consequence of thermal stress. Additionally, the study reveals a negative association between TAC and HSP levels (ranging from - 0.660 to - 0.820), emphasizing the role of antioxidants in mitigating heat stress. The findings of this research offer compelling support for the critical role that HSPs play in protecting cells from heat-induced damage. Additionally, the presence of higher levels of HSPs in regions with more severe climate conditions serves as evidence of camels' adaptation to heat stress. These findings emphasize the substantial impact of environmental factors on HSP production and further reinforce the crucial role of HSPs in bolstering the resilience of camels. Further research is needed to explore HSP expression and mechanisms to effectively manage and enhance camel resilience in extreme temperatures.

Background: Therapeutic strategies with calcitonin gene-related peptide (CGRP) or its receptor have been investigated, but there are few studies regarding the possible harmful effects of CGRP in other body organs. Objective: This study aimed to investigate the effect of intracerebroventricular (ICV) injection of CGRP on sex hormones and sperm quality in rats. Methods: Twelve male rats were divided into two groups (n=6 per group). The first group (control) rats were injected with 5 µl artificial cerebrospinal fluid intra-ICV; the second group rats, 5 µl (1.5 nmol) CGRP. The levels of luteinizing hormone (LH), follicle-stimulating hormone (FSH), and testosterone were measured. Epididymal sperms were used to determine the sperm parameters. Results: The levels of testosterone, LH and FSH in CGRP group was significantly lower than in artificial cerebrospinal fluid (ACSF) group (P<0.05). The concentration and motility of sperm in CGRP group was significantly lower than in ACSF group (P<0.05). In CGRP group live spermatozoa and intact acrosome significantly reduced compared to the ACSF group (P<0.05). In addition, in CGRP group dead spermatozoa and lose acrosome significantly increased compared to the ACSF group (P<0.05). Conclusion: ICV injection of CGRP may reduce sperm quality, probably through induction of an imbalance in FSH and LH production as well as testosterone.

Parasitic blood diseases (theileriosis, babesiosis, anaplasmosis, and trypanosomiasis) are common in regions where the distributions of the hosts, parasites, and vectors are convergent. They endanger animal production, and a few are also harmful to public health. The acute phase reaction (APR) is a complex, non-specific reaction that occurs in various events, including surgical trauma, infection, stress, inflammation, and neoplasia. To understand pathogenesis, we must study APR effects and acute phase proteins (APPs) alterations in naturally occurring and experimental infections. The elevation of haptoglobin (Hp), Serum amyloid A (SAA), and fibrinogen concentrations was markedly significant in bovine and ovine theileriosis. Hp, SAA, ceruloplasmin, and fibrinogen concentrations in anaplasmosis were dramatically elevated. A significant increase in SAA was observed in bovine babesiosis, while ovine babesiosis showed a significant rise in sialic acid levels. In cases of trypanosomiasis caused by T. vivax, there have been reports of elevated levels of Hp, complement C3, and antitrypsin. Improving our understanding of APR could result in more effective methods for diagnosis, treatment, control, and eradication of diseases. The article provides an overview of APPs alterations and other inflammation-related parameters (some cytokines, adenosine deaminase, and sialic acids) in parasitic blood diseases of ruminants.

This study aimed to investigate the effects of probiotic and yeast extract supplementation on the metabolic, immune, and oxidative status of Sannen goat kids during the weaning challenge. Forty goat kids were randomly assigned to four groups: a probiotic group (Pr) (basal diet + mixture of Bacillus subtilis, Bacillus lechiniformis, Streptococcus Thermophilis, and Enterococcus faecium), a yeast cell wall extract group (YC) (basal diet + Saccharomyces cerevisiae), a probiotic and yeast cell wall extract group (Pr + YC) (basal diet + mixture of probiotic and yeast cell wall extract), and a control group (basal diet). Treatments were administered 21 days prior to weaning (80 ± 2 days of life) until 21 days post-weaning except for the control group. Blood samples were collected at four different time points, including 21 days before weaning, 2 days post-weaning (weaning time), 7 days post-weaning, and 21 days post-weaning. Average levels of triiodothyronine, thyroxine, total protein (TP), albumin, globulin, blood urea nitrogen (BUN), total antioxidant capacity (TAC), serum adenosine deaminase (ADA), nitric oxide (NO), ferritin, glucose, cortisol, triglyceride, non-esterified fatty acid (NEFA), Β-hydroxybutyric acid (BHBA), and body weight were measured. The average levels of cortisol tended to be higher in the Pr group in comparison to the control group (P = 0.07) and the Pr + YC group (P = 0.10). NEFA was found to be higher and tended to be higher in the control group compared to the Pr + YC group (P > 0.001) and Pr group (P = 0.10), respectively. Additionally, the BHBA concentration was higher in the control group compared to the Pr group (P > 0.001). No differences were observed in the concentration of other measured parameters among the treatments. The concentration of cortisol tended to be higher (P = 0.10) at the weaning time as compared to the third sampling time. Furthermore, the concentration of TAC was observed to be higher (P > 0.01) at the weaning time in comparison to the third and fourth sampling times. The concentration of NO was higher (P > 0.01) at the third sampling time when compared to the first sampling time. A reduction in NEFA and BHBA levels may suggest an improvement in the metabolic status of the supplemented animals during the weaning challenge. However, supplementation with probiotics and yeast cell wall extract did not appear to have an effect on the oxidative status of the animals. The increase in TAC and NO levels observed during the weaning time may indicate an increase in oxidative stress during this period.

Piroplasmida is an order of economically important blood parasites, including Babesia, Theileria, and Cytauxzoon, transmitted to mammals by ticks. Oxidative stress is a state in which the balance between oxidants and antioxidants is disturbed so that antioxidants cannot compensate for the harmful effects of oxidants. Due to the high concentration of oxygen and hemoglobin in red blood cells (RBCs), these are among the first cells exposed to oxidative damage. When RBCs are exposed to oxidative stress, their hemoglobin is oxidized, and lipid peroxidation leads to membrane instability, deformation, hemolysis, and anemia. Oxidative stress has a fundamental role in the pathogenesis of these parasites. In the present review article, we collected studies on the oxidative stress caused by Piroplasmida in domestic animals. What is obtained from the total review of studies conducted on piroplasmosis revealed that in these infections, the host faces oxidative stress, and the resultant oxidative injury plays a fundamental role in their pathogenicity. Further studies are needed to be carried out on the more precise role of oxidative stress, the use of more sensitive diagnostic biomarkers, and the possible therapeutic role of antioxidant agents in piroplasmosis.

Trypanosoma species cause animal trypanosomiasis that infects many animals. Trypanosoma evansi is an organism that infects camels. There are many economic problems associated with this disease, including lower milk and meat yields and abortions. The purpose of the current survey was molecular study of the presence of Trypanosoma in dromedary camel blood in the south of Iran, and its effects on the hematologic, and some acute-phase protein changes. Blood samples were aseptically collected from the jugular vein of dromedary camels (n = 100; aged from 1 to 6 years) originating from Fars Province in EDTA-coated vacutainers. Genomic DNA from 100 µL of the whole blood was extracted and amplified using a PCR assay based on ITS1, 5.8S, and ITS2 ribosomal regions. Also, the PCR products obtained were sequenced. Moreover, the changes in hematological parameters and serum acute-phase proteins (serum amyloid A, alpha-1 acid glycoprotein, and haptoglobin) were measured. Among 100 tested blood, nine samples (9%, 95% CI: 4.2-16.4%) were found positive by the PCR assay. The phylogenetic tree and blast analysis showed four different genotypes closely related to the strains (accession numbers: JN896754 and JN896755) previously reported from dromedary camels in Yazd Province, center Iran. Based on hematological analysis, normocytic and normochromic anemia and lymphocytosis were detected in the PCR-positive cases compared with the negative group. Furthermore, alpha-1 acid glycoprotein was significantly increased in the positive cases. There was a substantial and positive relation between the number of lymphocytes, and the levels of alpha-1 acid glycoprotein and serum amyloid A in the blood (p = 0.045, r = 0.223 and p = 0.036, r = 0.234, respectively). A noticeable frequency of T. evansi infection was reported in dromedary camels in south Iran. This is the first report on the genetic diversity of T. evansi in this region. There was a significant association among Trypanosoma infection, lymphocytosis, and alpha-1 acid glycoprotein. Trypanosoma-positive camels had a significant decrease in hematocrit (HCT), hemoglobin (Hb), and red blood cell (RBC) values compared to the non-infected group. Further experimental studies are needed to elucidate the hematological and acute-phase protein alteration during a different phase of Trypanosoma spp. infection.

BACKGROUND: This research was designed to evaluate the effects of therapeutic ultrasound waves on ovarian germinal tissue and inflammatory cytokines (interleukin-6 (IL-6), IL1ß, tumor necrosis factor-α (TNF-α)), acute phase proteins (serum amyloid A (SAA), C reactive protein (CRP)) and oxidative stress (total antioxidant capacity (TAC), and malondialdehyde (MDA)) in dogs. Twenty-six clinically healthy adult mix-breed female dogs were aligned into three groups. Laparotomy was performed in control (n = 6) and treatment (T5, n = 10; T10, n = 10) groups. The ultrasonic exposure of ovaries in treatment groups was performed during laparotomy by round motions of the therapeutic ultrasonic transducer on both ovaries (1 MHz frequency, 1.5 W/cm2) for 5 min in the T5 group and for 10 min in the T10 group. Blood samples were collected from the jugular vein into a plain glass tube on days 0 (before laparotomy), 3, 6, and 9 after surgery. All control and treatment groups' dogs were ovariectomized for histological evaluation on day 60 after laparotomy or laparotomy + ultrasound exposure. RESULTS: Direct exposure of ovaries with therapeutic ultrasound waves induced inflammation and oxidative stress comparison with the control group. Histopathological evaluation of treated ovaries with ultrasound waves indicated a decreased number of primordial follicles (ovarian reserve) and oocyte preservation scores compared with ovaries in the control group. CONCLUSIONS: These changes may cause subfertility in the long term. It seems that inflammatory response and oxidative stress are factors in the permanent damage of ovarian tissue.

Nicotinamide adenine dinucleotide phosphate oxidase (NADPH oxidase), as a key factor in innate immunity, consists of several components, one of them is p40phox which is encoded by neutrophil cytosolic factor 4 (NCF4). Respiratory burst and reactive oxygen species (ROS) production are antimicrobial mechanisms associated with NADPH oxidase. This study evaluated the effects of g.18174 A > G and g.18270C > T single-nucleotide polymorphisms (SNP) in NCF4 on bovine mastitis and the respiratory burst capacity of neutrophils. SNPs of 160 dairy cattle were determined using a novel PCR-RFLP protocol by employing restriction enzymes, MboI and FokI. Also, the flow cytometry measured respiratory burst in 82 blood samples. Our results indicated that only g.18174 A > G SNP reduced the respiratory burst capacity. However, both SNPs were not significantly correlated with clinical mastitis. We concluded that g.18174 A > G decreases the function of NADPH oxidase. However, both SNPs were not significantly correlated with clinical mastitis.

The associations between early lactation changes in the body condition score (BCS) and mid-lactation levels of blood insulin-like Growth Factor-I (IGF-1), prolactin, insulin, nonesterified fatty acids (NEFA), milk production and some reproductive indices were investigated. Monthly milk yield and BCS were recorded. Blood samples were collected on days 60, 90, and 120 (±5 days) of lactation. Twelve and 17 cows showed BCS losses ≤0.75 and >0.75 on day 60, respectively. BCS losses ≤0.75 till day 60 after calving enhanced the IGF-I concentration on day 90 (57.8 ± 10.7 vs. 35.6 ± 17.8 ng/mL; p = .001). The differences in blood metabolites were prominent on day 90. Trends of insulin changes between day 60 BCS (p = .04) groups were different. Inverse correlations were detected between IGF-I and NEFA (-0.6, p = .001), IGF-I and milk production (-0.4, p = .05), NEFA and prolactin (-0.8, p < .001) on day 90. Higher percentages of cows with BCS loss >0.75 (day 60) showed their first heat after day 40 postpartum. In conclusion, irrespective of BCS at the time of breeding, the level of BCS loss during the first 60 days of lactation may affect further reproductive performance due to prolonged alterations of IGF-I, NEFA, prolactin and insulin levels. The extent of postpartum BCS loss may be a predictor of the metabolic status of cows during mid-lactation.

BACKGROUND: Ovariohysterectomy (OHE) induces inflammation and stress in female dogs. The anti-inflammatory effects of melatonin have been reported in several studies. OBJECTIVES: The goal of this study was to assess the effects of melatonin on the concentrations of melatonin, cortisol, serotonin, α-1-acid glycoprotein (AGP), serum amyloid A (SAA), c-reactive protein (CRP), interleukin-10 (IL-10), interleukin-8 (IL-8), interleukin-1ß (IL-1ß) and tumour necrosis factor-α (TNF-α) before and after OHE. METHODS: The total number of animals was 25 and aligned in 5 groups. Fifteen dogs were divided into three groups (n = 5): melatonin, melatonin+anaesthesia and melatonin+OHE and received melatonin (0.3 mg/kg, p.o.) on days -1, 0, 1, 2 and 3. Ten dogs were assigned to the control and OHE groups (n = 5) without melatonin treatment. OHE and anaesthesia were performed on day 0. Blood samples were obtained via jugular vein on days -1, 1, 3 and 5. RESULTS: Melatonin and serotonin concentrations significantly increased in the melatonin, melatonin+OHE and melatonin+anaesthesia groups compared with the control group, while cortisol concentration decreased in the melatonin+OHE group compared with the OHE group. The concentrations of acute-phase proteins (APPs) and inflammatory cytokines significantly increased after OHE. The CRP, SAA and IL-10 concentrations decreased significantly in the melatonin+OHE group compared with the OHE group. The concentrations of cortisol, APPs and proinflammatory cytokines increased significantly in the melatonin+anaesthesia group compared with the melatonin group. CONCLUSIONS: The oral administration of melatonin before and after OHE help controlling the high levels of inflammatory APPs, cytokines and cortisol induced by OHE in female dogs.

In all equids worldwide, Theileria equi and Babesia caballi are believed to be two important erythrocytic protozoa that cause equine piroplasmosis. In addition, it was recently discovered that Theileria haneyi is another potential equine piroplasmosis (EP) agent. Ixodid ticks are the major vectors of these parasites. Equine piroplasmosis is of international importance and affects enormously the equine industry. In this study, for the first time, molecular prevalence and genetic diversity of piroplasma parasites (T. equi and B. caballi) in horses from Fars province (south of Iran) were determined. Also, hematological alterations of naturally infected horses were analyzed. PCR positive horses showed anemia, thrombocytopenia, leukocytosis with a left shift of neutrophilia, and monocytosis. PCR results revealed that, from 133 blood samples of horses, 40 samples were positive (30.07%). The occurrence of T. equi in this area (30.07%) was more than the national average prevalence of T. equi (24.11%), but B. caballi prevalence in study area (0%) was less than the average of previous studies in Iran (5.47%). Our findings revealed that the T. equi was widespread in Fars province of Iran. PCR products of 18S rDNA and EMA-1 genes of T. equi strains were sequenced successfully. All 18S rDNA sequences collected in this experiment revealed 100% similarity together. According to the phylogenetic tree constructed using the 18S rDNA gene, Iranian T. equi is clustered with strains from Cuba (KY111762, KY111761) and USA (CP001669, JX177672). So, this could be concluded that T. equi studied in this research, and those strains are initiated from a common T. equi ancestor at an unknown time ago. Also, the phylogenetic tree based on EMA-1 gene demonstrated a genetically diverse population of Iranian T. equi strains (10 different genotypes). As EMA-1 is one of the most immunogenic antigens in this parasite, such variability could be a concern about the efficacy of T. equi vaccines. Finally, more studies on equine piroplasmosis in the provinces of the southern region of Iran are recommended to create a better vision of disease in this region.

Multiple sclerosis (MS) results from the destruction of myelin and focal inflammation. The study aimed to evaluate the effect of hydroalcoholic extract of Urtica dioica on oxidative stress, heat shock proteins, and brain histopathology in multiple sclerosis model. Sixty male C57BL/6 mice were divided into six groups of 10. Groups included positive control, negative control, and treatment groups with U. dioica extract at a dose of 50, 100, 200, and 400 mg/kg for 21 days (three times a week). The MS model was developed by a diet containing 0.2% cuprizone for 6 weeks. A section of brains was evaluated with Luxol Fast Blue staining and the other part evaluated with heat shock protein (HSP) kits 60 and 70, total antioxidant capacity (TAC), and malondialdehyde (MDA). In sections of corpus callosum, the highest amount of myelin was observed in the negative controls, while the use of cuprizone in the positive controls caused the destruction and reduction of myelin. The use of U. dioica extract in therapeutic groups except at a dose of 50 mg/kg could reduce myelin degradation to some extent and lead to remyelination. However, myelin levels in treatment groups were not significantly different from any of the negative and positive controls. Although HSP60 decreased in the treatment groups, there was no significant difference between the positive and negative controls. Treatment with this extract significantly reduced the amount of HSP70 compared with the positive controls. The decreased TAC and increased MDA in positive controls indicated oxidative stress, respectively. Furthermore, the extract led to an increase and decrease of TAC and MDA in the treatment groups, respectively. However, only the MDA level was significantly different from that of the positive controls. Therefore, the antioxidant effects of U. dioica extract could decrease cuprizone-induced oxidative stress and be effective in improving demyelination.

BACKGROUND: Although normal haematological and serum biochemical values for both pet and wild birds have been published, little information is available on the haematological and serum biochemical values in long-legged buzzards (Buteo rufinus). OBJECTIVES: This is the first study that aimed to define reference values of haematological, biochemical parameters, and protein electrophoretic fractions of long-legged buzzards in Iran. METHODS: Blood samples were collected from 30 clinically healthy adult long-legged buzzards of both sexes. Hematological, biochemical parameters, and protein electrophoretic fractions were measured. The mean and standard deviations were calculated. RESULTS: Mean values for red blood cells, packed cell volume, haemoglobin, and white blood cells were 2.72 ± 0.60 ×106 /µl, 39.10 ± 3.70%, 13.45 ± 1.30 g/dl, and 3.92 ± 1.39 ×103 /µl, respectively. Mean values for biochemistry parameters were total protein 4.46 ± 1.27 g/dl, albumin 1.78 ± 0.55 g/dl, creatinine 0.54 ± 0.22 mg/dl, uric acid 7.81 ± 2.89 mg/dl, calcium 9.63 ± 2.22 mg/dl, phosphorus 4.31 ± 1.00 mg/dl, glucose 398.87 ± 96.90 mg/dl, blood urea nitrogen 10.46 ± 3.85 mg/dl, alkaline phosphatase 127.01 ± 1.46 IU/L, aspartate aminotransferase 262.22 ± 116.30 IU/L, and alanine aminotransferase 56.63 ± 27.85 IU/L. Mean values for serum protein fractions included pre-albumin, albumin, α-1 globulin, α-2 globulin, ß- globulin, and ϒ-globulin was 0.20 ± 0.09, 2.35 ± 0.67, 0.28 ± 0.13, 0.32 ± 0.07, 0.62 ± 0.24, and 0.68 ± 0.53 g/dl, respectively. CONCLUSION: The reference data presented in this study can be used as health assessment values for veterinary laboratories and clinicians when developing release criteria for rehabilitated long-legged buzzards.

The aim of this study was to investigate gonadectomy stress, steroid hormones and serotonin in male dogs treated with melatonin before gonadectomy. Twenty-five mixed breed adult dogs were divided into five equal groups. The melatonin and melatonin + gonadectomized groups received melatonin treatment (3 mg/10 Kg, PO, TID) the day before gonadectomy; the gonadectomized and anaesthesia groups did not receive melatonin; and the control group just received the melatonin vehicle. Blood sampling was performed before melatonin administration (day -1) and on days 0 (gonadectomy), 1, 3 and 6 after gonadectomy. Superoxide dismutase and glutathione peroxidase concentrations decreased significantly in gonadectomized dogs compared with dogs treated with melatonin before gonadectomy and intact dogs. Gonadectomy led to a significant decrease in catalase concentration in gonadectomized dogs compared with other study groups. Malondialdehyde levels increased significantly in gonadectomized dogs compared with other groups. Melatonin administration before gonadectomy led to decreased malondialdehyde concentration in gonadectomized and intact dogs compared to the control group. Cortisol concentration increased significantly in gonadectomized dogs compared to the control dogs. Serotonin levels decreased in gonadectomized dogs, but melatonin treatment increased serotonin concentration in gonadectomized and intact dogs. Melatonin treatment before gonadectomy suppressed oxidative stress and the cortisol but increased serotonin level.

BACKGROUND: Major histocompatibility complex (MHC) is the best characterised genetic region associated with adaptive immune responses, including humoral and cell-mediated immunities. OBJECTIVES: In this study, the association of MHC class II alleles with inflammatory cytokines and acute-phase proteins was evaluated in sheep population. METHODS: Allelic diversity of second exon of ovine DRB1 locus (Ovar-DRB1.2) was determined in 100 indigenous Iranian Lori-Bakhtiari fat-tailed sheep using restriction fragment length polymorphism and direct sequencing methods. The association of DRB1.2 alleles with inflammatory cytokines (interleukin-1ß, IL-1ß; IL-6 and tumour necrosis factor-α) and acute-phase proteins (serum amyloid A, alpha-1-acid glycoprotein and haptoglobin) was examined using generalised linear model and multivariate regression analysis. RESULTS: Seven distinct RsaI restriction patterns and fourteen alleles were identified in this population. Allele DRB1*2101 showed a negative influence on the IL-6 response and was associated with lower serum level of IL-6. DRB1.2 heterozygous individuals also showed higher haptoglobin concentration than homozygotes. CONCLUSIONS: These results provide additional support for the association between Ovar-DRB1 alleles and regulation of immune responses in sheep population. Description of MHC polymorphism and its role in the controlling of immune responses will increase our understanding of host-pathogen interactions, and ultimately facilitate the selection of disease-resistant flocks in genetic breeding programs.

This study was conducted to evaluate the effect of maternal exposure to TiO2 nanoparticles on the pain response in offspring mice. 30 female mice with a mean ± SD weight of 30 ± 5 g were randomly divided into three groups: the control group (group 1) received only the basal diet; the sham group (group 2) received saline plus as a carrier (100 µL/mice) subcutaneously on days 3, 7, 10, and 14 post-mating; and the test group (group 3) received 100 µL/mice TNPs subcutaneously on days 3, 7, 10, and 14 post-mating. Offspring were divided into 6 groups 21 days after birth and underwent formalin test. Blood samples were taken to evaluate possible oxidative changes in total antioxidant capacity (TAC) and malondialdehyde (MDA). Exposure to TNPs significantly (p < 0.05) decreased pain perception. Except for a significant difference between the sham group and the control group, MDA and TAC were not significantly different among the studied groups. Injection of TNPs to pregnant mice would affect the pain perception in their offspring. This may be attributable to the ability of these particles to pass through the placenta to produce free radicals.

The aim of this study was to evaluate the changes of ghrelin, microminerals, antioxidants, and vitamins A, E and C levels during different metabolic periods in high yielding Saanen goats subjected to heat stress. Twenty clinically and paraclinically healthy, high yielding and multiparous goats with an average age of 3 ± 0.5 years and pregnant with a single fetus were included in this study. Sampling was performed at three different physiologic periods: non-pregnancy non-lactation (P1), four-month gestation (P2), and first month of lactation (P3). In this study, the ambient temperature ranged from 19 to 42 °C and relative humidity ranged from 14 to 19% during the hot months. Serum concentrations of ghrelin, glucose (Glu), superoxide dismutase (SOD), glutathione peroxidase (GPx), vitamins (A, E and C) and microminerals (selenium, manganese, cobalt, iron, copper and zinc) were measured. Mean raw milk yield of the goats per day at the first month of lactation was 2.34 ± 0.2 kg. Concentration of ghrelin at P1 was significantly lower than P2 and P3 (P < 0.05). Glucose levels were significantly lower at P3 compared with P1 and P2 (P < 0.05). There were no significant correlations between ghrelin and Glu at different periods. Concentrations of selenium and manganese were significantly higher at P3 compared with P2 and were significantly higher at P2 compared with P1. Values of copper at P2 were significantly higher than P1 and P3 (P < 0.05). Zinc levels were significantly higher at P1 compared with P2 and P3 (P < 0.05). Values of antioxidants and vitamins were significantly lower at P3 compared with P2. It is concluded that high yielding Saanen goats may suffer from hormonal and metabolic disturbances, oxidative stress and micromineral deficiencies during late gestation and the first month of lactation especially when they are subjected to heat stress.

INTRODUCTION: Gastric ulcer is a multifaceted process and is usually caused by mucosal damage. Herbal medicines have received much attention considering the side effects of chemical drugs. Nowadays, the use of herbal medicines has received much attention considering the side effects of chemical drugs. Quercus brantii Lindl, Cirsium vulgare (Savi) Ten, and Falcaria vulgaris Bernh are plants used as traditional phytomedicine for gastric ulcer diseases. AIM OF THE STUDY: This study was aimed to investigate the protective effects of hydroalcoholic extracts of these herbs on ethanol-induced gastric ulceration, in addition, to investigate the antioxidant, anti-inflammatory, and gene expression. MATERIALS AND METHODS: Thirty Sprague Dawley rats, (200-250 g), were divided into six groups: Control: intact animals; sham: gavaged with distilled water (14 days); negative control: gavaged with 20 mg/kg of omeprazole (14 days); experimental groups I, II, and III: gavaged with 500 mg/kg of the extract of Falcaria vulgaris, Quercus brantii, and Cirsium vulgare, respectively, (14 days). The number of ulcers and pathological parameters were assessed. The serum superoxide dismutase, catalase, glutathione peroxidase, malondialdehyde, total antioxidant capacity, albumin, total protein, haptoglobin, alpha-1-acid glycoprotein, total globulin, alpha-2-macroglobulin, C-fos, C-myc, and Caspase-9 were measured by ELISA and RT-PCR. RESULTS: The extracts significantly reduced gastric ulcer (52.33%). The results showed that the Quercus brantii extract was more effective. There were significant differences between the serum levels of alpha-1-acid glycoprotein and those of alpha-2-macroglobulin. Also, there was a significant difference in the serum level of antioxidant parameters. Changes in the expression of the genes also confirmed the results suggested by other parameters. The expression levels of C-fos, C-myc, and caspase-9 were decreased, but the Bcl-2 expression increased. CONCLUSION: The hydro-alcoholic extracts revealed various protection and noticeable change in the expression of caspase-9, C-myc, C-fos, and Bcl-2 genes in rats.
